Abstract
1α-25-dihydroxyvitamin D3 exerts antitumour effects in vitro and in vivo, but its potential therapeutic value is limited by its strong calcemic effect. New vitamin D analogues with a more favourable therapeutic profile are therefore under development.
Vitamin D analog EB-1089 is approximately 100 times more potent than 1,25-(OH)2D3 in inhibition of U937 human histiocytic lymphoma cells proliferation, and 50 times in human breast cancer cell line MCF-7.
In rats with mammary tumors induced with nitrosomethylurea, oral treatment with 0.5μg/kg/day EB-1089 for four weeks resulted in significant inhibition of tumor growth without changes in serum calcium levels.
